To address these inconsistencies, researchers conducted a comprehensive meta-analysis to evaluate all available evidence regarding magnesium's effectiveness in reducing depression symptoms among adults diagnosed with depressive disorders.

This study involved an extensive systematic search of online medical databases, incorporating all relevant terminology through July 2023. The analysis included only randomized controlled trials that compared magnesium supplementation against placebo treatments for measuring depression outcomes.

The final analysis encompassed seven clinical trials with a combined participant pool of 325 individuals, with average ages spanning 20 to 60 years. These studies generated eight distinct effect measurements. The meta-analysis revealed statistically significant improvements in depression scores following magnesium supplementation.

Nearly half of older adults experience insomnia, which includes difficulty falling asleep, early morning awakenings, or feeling unrefreshed upon waking. Aging is associated with several factors that increase the risk of insomnia, including changes in circadian rhythms, environmental and lifestyle changes, and reduced intake, absorption, retention, and utilization of nutrients.

Magnesium (Mg2+), a natural N-methyl-D-aspartic acid (NMDA) antagonist and GABA agonist, have an important role in regulating sleep. This study evaluates the effectiveness of magnesium supplementation in improving insomnia in elderly individuals.

A double-blind, randomized clinical trial was conducted with 46 elderly participants who were randomly assigned to receive either 500 mg of magnesium or a placebo daily for 8 weeks. The Insomnia Severity Index (ISI), physical activity levels, and sleep logs were assessed at the beginning and end of the study. Dietary intake of magnesium, calcium, potassium, caffeine, carbohydrates, and total calories was measured using a 24-hour recall over three days.

Anthropometric variables were recorded to control for confounding factors. Blood samples were collected at baseline and after the intervention to analyze serum levels of magnesium, renin, melatonin, and cortisol. Data were analyzed using SPSS version 19, and P values less than 0.05 were considered statistically significant.

Compared to the placebo group, the magnesium group showed significant improvements in sleep duration, sleep efficiency, and increases in serum levels of renin and melatonin. ISI scores, sleep onset latency, and cortisol levels decreased in the magnesium group. There were also improvements in early morning awakening and serum magnesium levels.

This study shows that magnesium supplementation may improve insomnia in elderly individuals by improving subjective measures such as ISI scores, sleep efficiency, sleep onset latency, and early morning awakening, along with objective indicators like serum levels of renin, melatonin, and cortisol.
