Participants were randomly assigned to receive a placebo, or 300 mg, 600 mg, or 900 mg of NMN once daily.
The primary goal was to evaluate whether NMN increases blood NAD⁺ levels in a dose-dependent manner. Then it assessed the safety, tolerability, and clinical efficacy, including physical performance (via six-minute walk test), biological age (using Aging.Ai 3.0), insulin resistance (HOMA-IR), and self-reported health status (SF-36 questionnaire).
The results showed that all NMN groups showed significant increases in blood NAD⁺ levels at both day 30 and day 60 (p ≤ 0.001), with the highest levels seen in the 600 mg and 900 mg groups. Also, the six-minute walking distance improved significantly in all NMN-treated groups compared to placebo at both time points (p < 0.01), with the best results observed in the 600 mg and 900 mg groups. The biological age increased in the placebo group but remained stable in all NMN-treated groups, resulting in a significant difference between the two (p < 0.05).
