        AdvaCare’s Mycoplasma Test Kit is specially designed to detect the presence of mycoplasma contamination biological materials in cell cultures. It is known that the detection of mycoplasma by direct culture procedure is time-consuming and some mycoplasma species are hard to cultivate. By employing Polymerase Chain Reaction (PCR) testing to amplify the conserved 16S ribosomal RNA coding region within the Mycoplasma genome, our mycoplasma test kit aims to provide an extensive, highly sensitive, quick and efficient detection method.

        The PCR technology featured in AdvaCare’s mycoplasma test kit is easy to use, convenient and fast as the test results are obtained in less than 3 hours. In addition to this, our test kit is highly sensitive and can detect as little as 2 to 5 femtograms of Mycoplasma DNA in as little as 100 µL of test sample supernatant. A positive control is included with our test kit to enable the validation of the PCR amplification process and to confirm the size of the PCR product obtained in samples.

        AdvaCare’s mycoplasma test kits feature an internal control that successfully eliminates potential false negatives linked with PCR inhibitors and are manufactured following strict GMP, CE, ISO and USFDA standards.

        Usage

        Ensure that you wear gloves, masks, and a lab coat with tight-fitting cuffs during the procedure. Bring all reagents to room temperature before use.

        • Prepare reagents and samples as instructed;
        • Wash the Hybridization plate 2 times with Wash Buffer. Remove excess wash buffer by decanting or aspirating. Invert the plate and blot against clean paper towels;
        • Add 50μL of diluted probes to the designated wells;
        • Add 150μL of positive control, sample diluent (negative control), or sample to the designated wells. Cover with plate sealer;
        • Apply the float collar to the hybridization plate and incubate the plate for 60 minutes in a 65°C water bath;
        • Remove unused microplate strips from the streptavidin plate frame, return them to the foil pouch containing the desiccant pack, and reseal;
        • Wash the streptavidin plate twice with wash buffer and remove excess wash buffer as described in step 2;
        • Transfer 150μL from each well of the hybridization plate to the washed streptavidin plate and apply a new plate sealer;
        • Incubate for 60 minutes at room temperature on a horizontal orbital shaker set at 500 ± 50 rpm;
        • Wash the streptavidin plate 4 times with wash buffer and remove excess wash buffer;
        • Add 200μL of the anti-digoxigenin conjugate to each well and cover with a new plate sealer;
        • Incubate for 60 minutes on a shaker at room temperature;
        • Wash the streptavidin plate 6 times with wash buffer and remove excess wash buffer;
        • Add 50μL of substrate solution to each well and cover with a new plate sealer;
        • Incubate for 60 minutes on the shaker at room temperature. DO NOT WASH;
        • Add 50μL of amplifier solution to each well and cover with a new plate sealer;
        • Incubate for 30 minutes on the shaker at room temperature. DO NOT WASH;
        • Add 50μL of stop solution to each well;
        • Determine the optical density of each well within 30 minutes, using a microplate reader set to 490nm. If wavelength correction is available, set to 650nm or 690nm. If wavelength correction is not available, subtract readings at 650nm or 690nm from the readings at 490nm. This subtraction will correct for optical imperfections in the plate. Readings made directly at 490nm without correction may be higher and less accurate.

        Precautions

        • When handling cell culture samples, appropriate precautions should be taken to prevent exposure to mycoplasma and other hazardous biological agents;
        • Do not use test if pouch is damaged;
        • Humidity and temperature can adversely affect results;
        • The used test should be discarded according to local regulations;
        • Always wear protective gloves, clothing, eye, and face protection. Ensure that you wash your hands thoroughly after handling chemical products.
