FDA packaging validation must demonstrate that a medical device’s packaging consistently protects the device, preserves required sterility or cleanliness, and remains suitable through processing, distribution, storage, and labeled shelf life. Validation depth depends on the device and packaging risks.
Answer
Under the FDA Quality Management System Regulation, manufacturers must control packaging as part of product realization and validate packaging processes when their results cannot be fully verified through subsequent inspection. Requirements should be defined from the device’s intended use, sterility, physical characteristics, distribution environment, and claimed shelf life.
For terminally sterilized devices, FDA-recognized ISO 11607-1 and ISO 11607-2 are widely used to address sterile-barrier-system requirements and packaging-process validation.
A validation program commonly includes:
• Packaging design and material qualification
• Compatibility with sterilization and device materials
• Sealing-process installation, operational, and performance qualification
• Seal-strength and package-integrity testing
• Distribution and transportation simulation
• Accelerated and real-time aging
• Maintenance of sterility through labeled shelf life
• Worst-case configurations and justified sample sizes
• Label and UDI legibility after processing and distribution
• Change control and revalidation criteria
Common test methods may include ASTM seal-strength, dye-penetration, bubble-emission, accelerated-aging, and distribution-testing standards. The selected methods must be scientifically justified; completing a standard test does not by itself validate the entire packaging system.
Nonsterile devices may not require sterile-barrier testing, but their packaging must still protect against damage, contamination, deterioration, and foreseeable distribution stresses. Validation records should document protocols, acceptance criteria, deviations, statistical rationale, results, approvals, and conclusions.
